Performing Enhanced Multiparameter Cell Imaging with Combined Fluorescence Lifetime Imaging Microscopy and Atomic Force Microscopy

چکیده

optically encoded information about processes in live cells. Atomic force microscopy, on the other hand, provides nanometer-resolved surface topography and mechanical information, and has recently been expanded to nanometerresolved live cell mechanical property mapping. The integration of the two advanced live cell imaging techniques into one tool, with the capability to acquire simultaneous nanometer-mechanical and optical/biochemical information promises to provide new insights into cell mechanics. This application note describes the integration, demonstrated combined performance, and overlaid data of a cellular model system on the MicroTime 200 from PicoQuant and Bruker’s BioScope CatalystTM Atomic Force Microscope (AFM), on the basis of an IX71/81 Inverted Optical Microscope with PiFoc focus control.

Correlated atomic force microscopy and fluorescence lifetime imaging of live bacterial cells.

We report on imaging living bacterial cells by using a correlated tapping-mode atomic force microscopy (AFM) and confocal fluorescence lifetime imaging microscopy (FLIM). For optimal imaging of Gram-negative Shewanella oneidensis MR-1 cells, we explored different methods of bacterial sample preparation, such as spreading the cells on poly-L-lysine coated surfaces or agarose gel coated surfaces....

Simultaneous single molecule atomic force and fluorescence lifetime imaging

The combination of atomic force microscopy (AFM) with single-molecule-sensitive confocal fluorescence microscopy enables a fascinating investigation into the structure, dynamics and interactions of single biomolecules or their assemblies. AFM reveals the structure of macromolecular complexes with nanometer resolution, while fluorescence can facilitate the identification of their constituent par...

Application of Probes in Live Cell Imaging

Live cell imaging (LCI) is the approach of non-invasively analyzing dynamic processes in living cells using state-of-the-art microscopy and computer vision techniques. LCI provides exciting and novel insights into cell biology. The present review summarized LCI research in recent years and detailed the role of probes in LCI. We discussed the basic principle and development in LCI technology, in...
